Background :
	Interleukin-4 （IL4） has a compact, globular fold （similar to other cytokines）, stabilised by 3 disulphide bonds. One half of the structure is dominated by a 4 alpha-helix bundle with a left-handed twist. The helices are anti-parallel, with 2 overhand connections, which fall into a 2-stranded anti-parallel beta-sheet. Interleukin （IL）-4, produced primarily by CD4（+） T cells is an immunomodulatory cytokine, which can inhibit the growth of tumour cells.IL-4 has been known as a Th2 cytokine and can act on B cells, T cells, and monocytes.Interleukin （IL）-4 exhibits antitumor activity in rodent experimental gliomas, which is likely mediated by the actions of IL-4 on a variety of immune cells present in and around the tumor masses.
	Description :
	Recombinant Rat IL-4 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 126 amino acids and having a molecular mass of 14105 Dalton. Recombinant Rat IL-4 is purified by proprietary chromatographic techniques.
	Physical Appearance:
	Sterile Filtered White lyophilized （freeze-dried） powder.
	Formulation:
	Lyophilized from a concentrated （1mg/ml） solution in water containing no additives.
	Solubility:
	It is recommended to reconstitute the lyophilized IL-4 in sterile 18Mω-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
	Stability:
	Recombinant Rat IL-4 although stable at room temperature for 3 weeks, should be stored desiccated below -18⁰ C. Upon reconstitution, Interleukin-4 should be stored at 4⁰C between 2-7 days and for future use below -18⁰C. For long term storage it is recommended to add a carrier protein （0.1% HSA or BSA）. Please avoid freeze-thaw cycles.
	Purity:
	Greater than 98.0% as determined by: （a） Analysis by RP-HPLC. （b） Anion-exchange FPLC. （c） Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
	Amino acid sequence:
	The sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Gly-Cys Asn.
	Dimers and aggregates:
	Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
	Biological Activity:
	ProSpec’s Recombinant Rat IL-4 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant prolifiration of Con A activated rat spleen cells is < 1.5 ng/ml, corresponding to a Specific Activity of 6.6 x 10⁵ IU/mg.
	Endotoxin:
	Less than 0.1 ng/µg （IEU/µg） of Interleukin-4.
	Protein content:
	Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm using the absorbency value of 0.2 as the extinction coefficient for a 0.1% （1mg/ml） solution. This value is calculated by the PC GENE computer analysis program of protein sequences （InliGenetics）. 2. Analysis by RP-HPLC, using a standard solution of IL-4 as a Reference Standard.
	Usage:
